So I decided to give my approach on her skills in combat and also these suggestions will work a lot better with STD Beast (Pestilence).
So, the first thing you do is assess the defensive team and the turn order of your characters for better management of her skills. First off you start with her level 9 which gives her 3 bleed stacks and Blood Sacrifice buff which boosts her defense and attack.
There are 2 ways to start with Nico's skill set depending on the defenders; first, if the defense has full melee or one melee character that you consider can be troublesome, you start off with her Lv 2 Stick Around which gives Adhesive X, Constricted and Immobilized which comes in handy against a lot of melee heavy hitters and you basically make QS Juggernaut Rogue and many others obsolete for one round. The 2nd way is to start with the Fear me ability of her Lv 6 multifunction, which causes Cower and Intimidated. This option goes best with ranged attack characters or with characters that use quick actions first and you have the chance to stop their big hit in their tracks.
You should ALWAYS save her single target attacks and the More Dots and Rock n Roll abilities from her Lv 6 for when you are sure you can take out the entire defense or lower them to the point that they are no longer a threat. SAVE her You Suck ability from her Lv 6 because in most fights she's likely to be the last one standing due to her back from the brink passive and you want to use that on the last defending character to lower his attack and prevent future stat increases on his part.
Items/Weapons that work well with Nico are the Dark Sigil and the Chaos Shot SMG; Dark sigil applies dark void, burning and weak point, so you can pull off an early rock and roll for massive damage and it will be a guaranteed critical hit. For a higher survival rate of her and my other team mate i use the Hoarfrost mace, Sudden Support and Bruiser Empowered armor linked with 6 Spry and 2 Guardian e-iso and Surprising E-iso (never hurts to have a chance to stun someone). I've recently switched to the Generalist empowered armor socketed full reactive Chaotic (the silver and ShP version not the gold ones. if you can afford the gold ones then get those, they work better at max agent level) with the Inspiring e-iso that provides Morale boost to the entire team.
Best character team ups: 1 Sabretooth; he, like Nico, can resurrect once per turn, has an AoE potential stun and exploits attrition. and various chances to do a counter or follow up when an ally attacks with a single target attack due to his Lv 6 quick action, Berserker. 2 STD (Pestilence) Beast: although he can't resurrect, Beast's new alt suit gives him a different skill set and more useful effects. His Faustian bargain helps you keep the debuffs on the enemy and don't have to worry about them being removed and potentially losing Nico's worth in the game because of that. When there is a defending blaster, Tactician Beast is very handy, I always start with his Lv 6 quick action and follow with his Lv 1 on the blaster, generalizing him and gaining an extra turn to generalize and nanoplague whoever poses more of a threat on the field, or I use his Lv 9 on either a protector or a character that can do a lot of counter attacks or preemptive attacks (Quicksilver, Magneto, Rogue, Psylock etc).

Before using Nico Minoru's Lesser Arcana and Greater Arcana attacks, I wait till the opposing team uses Scroll of Angolob, The Light Fantastic and other debuff-removing actions. I rarely pair Beast with anyone but Nightcrawler because having him on the team is a double-edged sword, so Deadpool works a lot better for me. He has always been my main man and Happy to See You only helps to exploit the different effects Nico can inflict on enemies. Both can resurrect as well, proving to be an annoying team-up and offering much longer battles! You can use the Agent's Synthetic Cube primarily for its Improbability Field, helping add more “damage over time” effects.
